چکیده

The Pacific cod ( Gadus macrocephalus ) supports large commercial fisheries in the northern Pacific Ocean and the Bering Sea. Here we characterize 10 polymorphic microsatellite loci isolated from enriched genomic libraries. Loci were screened on a sample of 96 spawning adults. The average number of alleles per locus was 25.3 (range 12–44), with expected heterozygosities ( H E ) ranging from 0.54 to 0.97. No significant deviations of genotypic proportions from Hardy–Weinberg equilibrium or linkage equilibrium were detected. These markers will be used in future studies of population structure and mixed stock analysis of this important gadid species.

Microsatellite Isolation and Characterization in Pomegranate (Punica granatum L.)

Development of microsatellite markers has been an increasing trend in crop genetic studies because oftheir applicability in breeding programs. Here we report the development of inter simple sequencerepeat (SSRs) in pomegranate (Punica granatum L.) using an enrichment method that makes use of magneticbeads. Enriched genomic libraries with AG and ATG microsatellite motifs were c...

Characterization of di-, tri-, and tetranucleotide microsatellite markers with perfect repeats for Trypanosoma brucei and related species.

Trypanosoma brucei, unicellular parasites causing human sleeping sickness and animal nagana, have a great impact on the socio-economic environment of sub-Saharan Africa. The dynamics of the parasite are still poorly understood. We have characterized 14 polymorphic di-, tri-, and tetranucleotide microsatellite loci with perfect repeats (only one motif) exhibiting between 5 and 16 alleles in T. b...

Characterization of Novel Di-, Tri-, and Tetranucleotide Microsatellite Primers Suitable for Genotyping Various Plant Pathogenic Fungi with Special Emphasis on Fusaria and Mycospherella graminicola

The goals of this investigation were to identify and evaluate the use of polymorphic microsatellite marker (PMM) analysis for molecular typing of seventeen plant pathogenic fungi. Primers for di-, tri-, and tetranucleotide loci were designed directly from the recently published genomic sequence of Mycospherlla graminicola and Fusarium graminearum. A total of 20 new microsatellite primers as eas...

Application of Random Amplified Microsatellite Polymorphism (RAMP) in Prunus Characterization and Mapping

Random amplified microsatellite polymorphism (RAMP) is a PCR-based marker which uses a combination of two classes of markers: Simple sequence repeat (SSR) and Random amplified DNA polymorphism (RAPD) markers. RAMP has been demonstrated to be a potentially valuable molecular marker for the study of genetic relationships in cultivated plant species. The objective of this study was to optimize the...

Isolation and characterization of 13 new microsatellite markers in the triangle mussel (Hyriopsis cumingii)

Microsatellite enriched library of Hyriopsis cumingii was constructed according to the strong affinity between biotin and streptavidin. One hundred clones with foreign inserts were sequenced and 65 clones were screened for usable microsatellites, of which 36 were deemed unique, of sufficient length (more than 8 repeats) and possessed adequate flanking regions for primer designment. Among 36 pri...
